East Greenlandic polar bears, (a) Specimen age is estimated by counting the tooth cementum growth layers of the lower incisor (l3), and hence many skulls in the collection are missing this tooth, (b) Image of a 14-year-old male polar bear from Ittoqqortoormiit counted from a 14 fj thin layering cut stained with Toluidin blue using the method of Dietz et al.
The samples were stained using staining procedures specific for the desired attributes, as follows: Toluidin Blue (Feder and O'Brien, 1968) for the structural analysis; Coomassie Blue (Wetzel et al., 1989) for total protein content; and Periodic Acid Schiff (PAS, Gahan, 1984) for polyssacharides, acidic carbohydrates and neutral mucoproteins.
Thin sections, 5-6 micrometres, were cut using a microtome (Rotatory Microtome, Leica, RM 2265, Germany) and stained with Toluidin blue.
Histopathological diagnosis was made by observing mast cells showing metachromasia with toluidin blue in full-thickness skin biopsy (2).
There were no specific features with periodic acid-Schiff (PAS), toluidin blue (pH:1) and masson- trichrome histochemical stainings.
